Research Abstract |
OBJECTIVE : To determine the association of single nucleotide polymorphism in the several bone metabolism related gene and of an acquired factors with the spinal deformities in the cross sectional study. METHODS : A total of 432 women were invited to participate in the study. Physical examinations, thoraco-lumbar spinal radiographs, BMD measurement and blood samples were obtained from the subjects. We evaluated the presence of vertebral collapsed deformity, osteophytes, endplate sclerosis, disc narrowing, endplate sclerosis, and spinal scoliosis on the radiographs. Missense coding SNP for Q89R of the LRP5 gene was determined using the TaqMan PCR method. The polymorphisms of the other genes were determined as well. RESULTS : The value of age associated with BMD (r=0.35), osteophyte formation (r=0.44), and almost of spinal, degenerative factors. A significant correlation was observed between the weight and the BMD. The BMI correlated with disc narrowing and osteophyte formation. The value of osteophyte formation in farmer was significantly higher than that in other occupation. The value of disc narrowing in subjects with positive rheumatoid factors was significantly higher than that without the factor. We found that subjects without the R allele (QQ) of LRP5 gene had a significant lower osteophyte formation score than did subjects with R allele. The prevalence of scoliosis in the subject with the X-x allele of ER-alpha Xba I gene was 3.59 times higher than that with the other alleles. The mean number of collapsed vertebrae in the subject with the aa allele of 27-bp in the intron 4 of eNOS gene were tree or four times lager than that with the other alleles. CONCLUSIONS : We suggest that the genetic variation at several gene locus is associated with spinal deformities, and that the acquired factors affect the deformities as well.
