Research Abstract

脳組織におけるATPの細胞外放出をイメージングによって検証することが本研究の目的であった。昨年度、P2X_2受容体を発現させたHEK293T細胞がATP変化を検出し得るか検討したが、投与された外因性ATPに対する応答は長く持続的で、時間的な分解能が著しく低いこと、また、細胞間の発現効率の差のために十分な空間分解能が得られないことが判明した。以上より、細胞外代謝の速いATP濃度の変化を画像化するには、この方法は不適切であると結論した。そこで、本年度は新しい二つの試みを計画し実行した。第1は、超高感度光子検出カメラとluciferin-luciferase反応を用いて脳スライスにおける細胞外ATPを検出する試みである。海馬、大脳皮質、孤束核スライスにおいて電気刺激によるATP放出を画像化し得た。電気刺激による細胞外ATP濃度上昇は、(1)刺激電極の近傍に限局し、軸索終末近傍では観察されない、(2)テトロドトキシンによって消失しない、そして、(3)高K+濃度刺激によっては生じないことが明らかになった。これらは、脳スライスで電気刺激によってATPが放出されるとする現在までの報告の多くに重大な疑義を投げかける重要な結果である。第2は、バイオセンサーを用いた脳組織内局所的ATP濃度のリアルタイム計測である。同センサーは、2004年度までは開発国イギリス国内でしか使用できないという特許規定があったこと、また、開発者がヴェンチャー企業を設立し輸入手続に時間がかかったことなどから、研究代表者のもとに同センサーが届いたのは2005年1月であった。現在、同センサーを用いた脳スライスにおける細胞外ATP計測の実験を進行中である。以上より、本萌芽研究の目的は、当初の実験手法を変更したが、新たな方法を漸次採用することによって果たされ、萌芽段階ではない研究に進展しつつある。
