Research Abstract

サケ科魚類の回遊制御の分子機構を明らかにするためには,神経内分泌系に属する特定の神経核および神経下垂体のそれぞれに含まれるペプチド性の神経ホルモンの量を測定し,その動態を明らかにする必要がある.そのための超高感度アッセイ系を開発することを目的として研究を進めた.まずアッセイ系の開発を目指したペプチド性神経ホルモンは,サケ生殖腺刺激ホルモン放出ホルモン(gonadotropin-releasing hormone, GnRH)である.採用したアッセイ法の原理は,EIA法における発色剤の反応部分に分子生物学の研究でよく用いられるβガラクトシダーゼ(βGAL)の化学発光を利用することであった.アッセイ法の手順は,1)IgG抗体をコートしたマイクロプレートを用意する.2)マイクロプレート中で,標識ホルモンとしてビオチン化GnRHを用い,GnRH抗体存在下に,スタンダードあるいは試料中のsGnRHとの競合反応を行わせる.3)固層化されたGnRH抗体-ビオチン化GnRHの量を,アビジン-βGAL複合体を用いて定量する.検定は,モルキュラープローブ社のF-2905キットを用い,化学発光をフォトンカウンティングにより行う.今年度は,前年度のビオチン化GnRHの合成とそれに続くアビジン-βGAL複合体との結合の確認実験を踏まえて,フォトンカウンティングに適した基質の選択,および抗体のチェックを行い,10amol(amol=10^<-18>mol)の感度でスタンダードカーブを作成することができることを確認できた.しかし,感度を高めるためには化学発光の検出に時間をかける必要があり,必ずしも実用的といえる段階にまでは至らなかった.今後,手持ちの抗体より選択制の高い特異的なよい抗体を入手し,方法を改善する必要がある.
