Research Abstract

本研究では、糖鎖を厳密に認識するレクチン分子をモデルとして、そのキラル認識能についてD型タンパク質の合成およびキラル糖鎖との相互作用解析、複合体のX線構造解析により詳細な相互作用様式を明らかにすることを目的とした。そのため先ず魚類マアナゴ体表粘液に存在する2種のガレクチン(コンジェリンIおよびII)のうちコンジェリンIIについて、グリシンを除く全アミノ酸をD型とするタンパク質の化学合成を行い、Ac-Ser1-Gly35(C4 fragment), Glu36-Gly68(C3 fragment), Trp69-Gly104(C2 fragment), Glu105-Glu135(C1 fragment)の4つのセグメントに分割して合成を行った。すべてのフラグメントについての合成を終え、逆相高速液体クロマトグラフィー(HPLC)で精製した。チオエステル法よるフラグメント縮合法によりC1 fragmentとC2 fragmentの宿業をおこなった。フラグメントの不溶化により、効率的なカップリングおよび反応後の精製が困難であった。TofMSにより反応を確認したが、最終的なall-D型レクチン分子を大量に調製し、機能、特性を解析するまでには至らなかった。
本実験の副次的な研究成果として、コンジェリンIIをタグとした発現系構築へと発展させたところ、コンジェリンII自体がシャペロンとして働くこと、さらにシャペロンとの共発現系を構築したところ、これまで発現が非常に困難であった蛋白質(7組のジスルフィド結合を含む)およびその各種変異体を可溶化物として、また正しいフォールドで大量に発現させることに成功した。
