Research Abstract

我々は本研究で横紋筋肉腫で高発現する新規蛋白質ARPPを血液中で検出するためのELISAキットを開発した。捕捉抗体として我々が作製したマウス抗ヒトARPPモノクローナル抗体を96穴プレートに固相化し、ブロッキング後、固相化抗体と標準試料あるいは試料で第一反応した。標準試料にはGST/ヒトARPPリコンビナント融合タンパク0,0.16,0.8,4,20,100,500,2500ng/mlを、試料にはCOS-1細胞株とCHO細胞株の溶解液、およびヒトARPPを人為的に一過性強制発現させたCOS-1細胞株とCHO細胞株の細胞溶解液を用いた。反応後、検出抗体としてウサギ抗ヒトARPP抗体で第二反応、さらにHRP標識ヤギ抗ウサギ抗体で第三反応を行い、HRP活性をマイクロプレートリーダーで測定した。今回構築したヒトARPP ELISAのシステムでは、少なくとも0.16ng/mlのヒトARPPを検出する事が可能であった。また、ヒトARPPを一過性に強制発現させたCOS-1細胞株およびCHO細胞株の細胞溶解液のみにHRP活性がみられ、ヒトARPP特異的に反応することが証明された。
