Research Abstract

現在広く行われているプラスミドレベルの遺伝子断片の導入では、トランスジーンの挿入部位周辺の遺伝子発現の影響を強く受けること、ベクターの性質上、十分な発現制御領域をカバーできないことなどの問題点があり、しばしば導入遺伝子についての正確な情報を得ることができない。そこで本研究では、十分な発現制御領域をカバーできる細菌人工染色体(BAC)DNAを用いたトランスジェニックマウスを作製して、疾患感受性遺伝子を同定を行った。これまでの連鎖解析により、自己免疫疾患感受性の一つの候補遺伝子として想定されたCD72遺伝子が、疾患感受性遺伝子として機能しうるかどうかを明らかにするために、自己免疫疾患を発症するMRL/1prマウスに、抵抗性の多型を有するCD72 BAC DNAを遺伝子導入した。2系統の抵抗性多型のCD72BAC DNAを有したMRL/1prマウスと、コントロールとしてMRL/1prマウスと同様の感受性多型のCD72 BAC DNAを有したMRL/1prマウスを樹立した。これらのマウスを解析した結果、抵抗性多型BAC DNAを有するマウスでは、感受性多型のBAC DNAを有するマウスと比較して、寿命が著明に延長し、腎炎の改善を認めた。これらのマウスでの導入CD72の発現を解析したところ、正常のCD72の発現分布を示した。以上の解析結果から、連鎖解析によって同定されたCD72が実際に疾患感受性を決定していること、さらには導入したBAC DNAが生理的な発現を十分カバーできる領域を有していることが明らかとなった。従って、本研究によってBAC DNAを用いた、疾患感受性のスクリーニング方法が確立できたと考えられる。今後、これまで連鎖解析によって同定された他の疾患感受性遺伝子について、順次解析を行いたい。
