Research Abstract

本年度は、β_2-ミクログロブリン(β_2-m)アミロイド線維伸張反応のin vitro解析系(チオフラビンT法)を用いて、線維形成の分子機構とその阻害分子を解析する(論文1-4)とともに、アミロイド線維伸張反応の新しい解析系として、水晶発振子マイクロバランス法(Affinix Q)を用いる方法を検討した。これは水晶発振子センサーチップに断片化されたβ_2-mアミロイド線維を固定化し、それに対してリコンビナントβ_2-mを反応させて起こる重合反応を、水晶発振子の振動数変化として検知する系である。この系は、微量な試料を用いて定性的・定量的結合反応を高感度に評価できる利点がある。今年度、私たちは、その系におけるβ_2-mアミロイド線維形成反応および反応阻害実験の至適条件の設定を行った。
さらに今年度、私たちは、β_2-mを細胞内に取り込み代謝するエンドサイトーシス受容体メガリンの分子機能に検討を加え、advanced glycation endproducts(AGE)との直接の結合性を、水晶発振子マイクロバランス法を用いて証明した(論文5)。さらに、メガリンが肝型脂肪酸結合蛋白(liver-type fatty acid binding protein, LFABP)と結合し、その細胞内取り込みと代謝に関わることを明らかにした(論文6)。このような多面的な機能を有するメガリン分子の作用メカニズムを明らかにすることが、β_2-mアミロイド線維形成の阻害に有益であると考えられる。そこで私たちは、メガリンの巨大遺伝子を細胞内で発現させ、その機能ドメインを同定する研究を行った。既に、全長遺伝子を一過性に発現・機能させることに成功しており、部分的な遺伝子発現を行った場合との比較を行うとともに、定常的発現細胞の樹立を進めている。
