Research Abstract

ステロイドパルス療法の作用分子機構の解明を目的として、グルココルチコイド受容体(GR)の核内コンパートメント(foci)とAP-1クラスター形成との関係を高精細三次元画像解析システム(ZEISS-焦点レーザー顕微鏡/TRIグラフィックプログラム)を用いて解析した。また、ステロイド治療の際に臨床上しばしば遭遇するステロイド治療の二次無効について、GR foci形成の面から解析した。
1.GR fociとAP-1クラスター形成の相互作用の検討:GRはリガンドであるdexamethasone (Dex)を添加すると、核内に約300個のクラスター(foci)を形成し、このfoci形成とGRの転写活性化能は相関する。AP-1(fos/jun)と蛍光蛋白(GFP)のキメラ蛋白(AP-1-GFP)はこのGR foci形成を抑制した。また、逆にGRはAP-1クラスター形成を抑制し、両者は核内の発現レベルにより互いを抑制する平衡関係が存在することが明らかになった。
2.GR foci形成に対する大量グルココルチコイドの効果の検討:培養細胞(COS)にGR-GFPをトランスフェクションし、合成ステロイド(Dex10^<-9>〜10^<-6>Mあるいはメチルプレドニゾロン10^<-9>〜10^<-6>M)で処理すると核内にfociが形成されるが、24時間以上〜数日後はこのfoci形成が抑制される。この崩壊の程度は合成ステロイドの濃度が高いほど顕著であった。また、この崩壊はプロテアゾーム阻害剤で抑制された。この結果はGR foci形成が形成と蛋白分解の平衡の上に成立しており、大量グルココルチコイドは分解を促進していることが示唆された。
