Research Abstract

けいれん性障害の責任遺伝子・感受性遺伝子のリバースゲノミクス的スクリーニングとして、既に作出に成功している、AP3Bのμ3Bサブユニットをコードする遺伝子のノックアウトマウス(AP3B KO)・イノシトール3リン酸とGABA受容体γサブユニットの輸送蛋白をコードするPRIP遺伝子のノックアウトマウス(PRIP KO)に機能解析を進めてきたが、AP3B KOの自発性痙攣出現頻度は80%以上であったが、PRIP KOは自発性痙攣発現頻度が5%程度と低かったが致死性痙攣であった。この両遺伝子のダブルノックアウトマウス(DKO)の作出に成功し、致死性痙攣は無く、発作発現頻度はAP3B KOと同等の80%程度であった。また、PRIP KOは抗てんかん薬として一般的に用いられている、ベンゾジアゼピン系薬物(BZP)に対する感受性は著しく低下しており、DKOはBZP抵抗性のてんかんモデル動物となる可能性を示唆する結果が得られた。
