Research Abstract

クロモグラニンAは中枢神経系において広範に分布しているが、多様な神経伝達物質と共存してその遊離に関与していると同時に、ミクログリアの活性化に関与して神経細胞のアポトーシスを誘導していることが知られている。さらに、近年、クロモグラニンAの量が統合失調症の死後脳と脳脊髄液において減少していることが報告され、統合失調症の病態に関与しているのではないかと考えられている。そこで、クロモグラニンAを候補遺伝子として関連解析を行った。その結果、rs9658635SNPが有意な関連が見いだされた。双極性障害および覚醒剤使用障害サンプルを用いて関連解析を行ったが、クロモグラニンA遺伝子とこれら精神障害とは関連が見いだせなかった。したがって、クロモグラニンA遺伝子は統合失調症と特異的に関連していると考えられる。
また、細胞生物学検討として、マウスミクログリア細胞株Ra2および6-3におけるセロトニン(5-HT)受容体発現を確認した。まず、サイトセンサーシステムを用いて、ミクログリア細胞株を5-HTで刺激し、細胞外pH変化を検討した。その結果、serum free条件下で長期間培養すると、Ra2細胞株は5-HT刺激によって、弱いextracellular acidificationを起こすことが明らかになった。一方、カルシウムイメージングシステムを用いて細胞内カルシウム濃度変化を測定すると、Ra2細胞株において5-HT刺激による細胞内Ca^<2+>のオシレーションが観察されたが、6-3細胞株では5-HT反応は観察できなかった。
統合失調症の病勢の進行には神経変性過程が関与していることが予測されており、本研究結果から、5-HT系を介してミクログリアが抗精神病薬等向精神薬の作用点となっている可能性が推測される。
