Research Abstract

1 マウスに腫瘍細胞FSa-IIに蛍光蛋白enhanced green fluorescent protein (EGFP)を遺伝子導入して細胞株FSa-IIFを作製した。FSa-IIFは、Scidマウス、C3Hマウスに容易に移植腫瘍を形成した。
2 細胞株FSa-IIを用いて加温後のHSP70発現をインビトロレベルでウェスタンブロットを用いて検討した。45度までの加温では、加温後細胞内にHSP70が増加した。
3 新開発した三次元温度分布表示用ファントムを用いて、刺入型アプリケータによる0.5wattでのRF加温を行った。刺入型アプリケータ近傍には45度以上の高温域が出現し、その周囲には45度以下の低温域が存在した。
4 C3Hマウスに細胞株FSa-IIを移植し、刺入型アプリケータによる0.5watt、5分間のRF加温を行った。TG50値を算出できるマウス数での検討はいまだ完了していないが、無処置、RF加温単独、樹状細胞移植単独、樹状細胞移植併用RF加温の群に分けて処理後腫瘍の大きさを観察し、無処置、樹状細胞移植単独と比して、RF加温群、さらに樹状細胞移植RF加温併用では強い増殖遅延傾向が観察され、ひきつづきインビボでの検討を続ける予定である。
