癌抑制遺伝子p53産物のC末端ペプチドによる放射線・温熱癌治療増感

Research Abstract

目的
p53を標的とした癌治療にはp53遺伝子治療が脚光を浴びているが、変異型p53は正常型p53に対して優性に働くので、変異型p53,遺伝子を持つ患者にはp53遺伝子の遺伝子治療には困難が伴う。ここで申請している人工的に合成したp53タンパク質のC末端(アミノ酸残基361-382と353-374)のみからなるペプチド治療は変異型p53タンパク質にアポトーシス誘導能のみを回復させ、変異型p53患者においても放射線・温熱・制癌剤それぞれの治療に効率の良いアポトーシス誘導をねらっており、より効率の高い癌治療法の開発を目的としている。我々はヒト癌細胞の培養細胞系において、p53C末端ペプチドで変異型p53をもつ癌細胞をあらかじめ処理をすれば、癌細胞が放射線/温熱に感受性になること、p53C末端ペプチドは変異型p53タンパク質にアポトーシス誘導能を回復させる働きがあることをこれまでに明らかにした。本研究ではさらにヌードマウスにヒト癌細胞を移植した系においてもp53C末端ペプチドに増殖抑制効果があるかを実証することを目的とした。
方法
p53を欠損した非小細胞肺癌細胞H1299に変異型p53を導入したH1299/248細胞(変異型p53)またはneoコントロールベクターのみを導入したH1299/neo細胞(p53欠損)をヌードマウス(Balb/cAJcl-nl)の大腿皮下に移植し、p53C末端ペプチド(40μg/mlを腫瘍体積の1/2量)を局所投与した。経日的に腫瘍径を計測し、腫瘍重量を算出した。
成果
p53C末端ペプチド処理によって変異型p53腫瘍はp53欠損腫瘍に比べ明確な増殖遅延を示した。培養細胞を用いた実験と同様にヌードマウス移植癌実験においてもp53C末端ペプチドの分子シャペロン効果が確認された。この研究成果により、変異型p53遺伝子を保有する癌患者の場合にも、より効率の高い治療法が可能となり得ることが示唆された。
