Research Abstract

1.培養細胞へのCDX-2強制発現ベクター導入を目指し、ヒト正常大腸組織から得たcDNAをもとにCDX2遺伝子のサブクローニングを行ったが、正常食道上皮細胞の培養に必要な牛下垂体抽出物を含む特殊な培養液が狂牛病対策によって入手が困難となり、実験の中断を余儀なくされた。
2.Barrett食道の発生機序を生体を用いて解明するため、(1)CAG promoter-LoxP CAT LoxP-lacZ-CDX2 transgenic mouse(2)CK14promoter-Cre transgenic mouseの交配によるCK14-CDX-2 transgenic mouseの作成を進めている。現在までに
(1)K14 promoter constructをProfessor Elaine Fuchs (Laboratory of Mammalian Cell Biology and Development, The Rockefeller University)(2)Mouse CDX-2 cDNAを青木正博博士(京都大学医学研究科遺伝薬理学教室)(3)CAG promoter-LoxP CAT LoxP-lacZ-plasmidを宮崎純一教授(大阪大学大学院医学系研究科 未来医療開発専攻 幹細胞制御分野)(4)nls.Cre ERTM cassetteをProfessor Mark A.Magnuson (Department of Molecular Physiology and Biophysics, Vanderbilt University School of Medicine)よりそれぞれ供与を受けトランスジーンコンストラクトを作成中である。コンストラクト作成ののち培養細胞における発現確認を行い、トランスジェニックマウスの作成を行う予定である。
