膵島移植における分離膵島の二層法による機能回復に関する研究

Research Abstract

膵島移植において、膵島分離後24〜48時間の培養(例えばRPMI,37℃で培養)を行ったのちに移植することを推奨する報告が多い。我々はこの移植前の膵島に対する培養に二層法を組み合わせ、成績向上を目指した。Wister ratの膵島を分離後、種々の条件で24時間の培養あるいは保存を行い、膵島のviability評価をin vitroで行った。1群:培養・保存なし(control)、2群:RPMI,37℃で培養、3群:二層法(RPMI/PFC)37℃で培養、4群:二層法(RPMI/PFC)4℃で培養、5群:UW,4℃で保存、6群:二層法(UW/PFC)、4℃で保存。Stimulation Indicesはそれぞれ1.3,2.6,3.7,1.2,1.4,2.4で2、3、6群ではインスリン刺激に対する膵島の反応性が良好であった。これら3群における膵島をFDA/PI染色でそのviabilityを比較した結果、2、3、6群でそれぞれ88%,92%,76%で、有意差を認めないものの、二層法(RPMI/PFC)37℃が最も良好な結果を示した。以上より、分離膵島の培養に二層法を応用する手法の有用性を証明した(Hepato-Gastroenterol, in press)。
