Research Abstract

異なる2波長での蛍光強度を測定しその比(レシオ)を検出するレシオ測定法は,バイオイメージングを行う際に特に有利な測定法である.レシオ測定を行うためには,標的分子との相互作用で励起波長ないし蛍光波長がシフトする特性をもつ蛍光プローブが必要となる.本年度は,蛍光共鳴エネルギー移動(FRET)を基本原理としたレシオプローブ設計法の確立を行った.空間を介したエネルギー移動であるFRETの効率は,Forster式に従って変化する.このうち,重なり積分:Jに着目し,プローブ分子をデザインした.フルオレセインが吸収特性の大きく異なる2つのコンフォメーションをとることに着目して,重なり積分変化をスイッチとする設計法を新規に考案した.この原理に基づき,リン酸基を導入した1__-をチロシンホスファターゼ(PTP)レシオプローブとして設計・合成した.1__-は水溶液中で450nm付近のクマリン蛍光を示したが,PTP添加するとクマリン蛍光が減少し515nm付近のフルオレセイン蛍光が増大した.1__-を生細胞に導入しレシオイメージングを行った結果,経時的にレシオ(アクセプター蛍光/ドナー蛍光)の増大が観測された.次に,1__-を用いたイメージングで正常細胞の接触阻止とPTP活性の関連について解析することを試みた.接触阻止を起こすマウス筋芽細胞C2C12を用いて細胞密度とPTP活性との関係を調べたところ,周囲がすべて細胞で埋まっているDensoの状態において,PTP活性の増大が観測された.一方,接触阻止能を欠損しているHeLaでは,細胞密度によるPTP活性の変化は見られなかった.よって,生きた細胞からPTP活性を直接検出することで,接触阻止が起こる際にPTP活性が増大することを示した.また,阻害剤を用いた検討の結果,アラキドン酸代謝経路より産生されるROSがPTP活性の調節因子となっていることが示唆された.
