Research Abstract

水チャネルであるアクアポリン(AQP)は、様々な臓器に発現し、水の移動の調節、細胞容積の調節、ホルンモンの分泌などに関与している。脳浮腫は頭部外傷、脳血管障害、脳腫瘍など様々な病態に随伴して発症し、しばしば致命的となる。その病態は、アストロサイト(Ast)の膨化(水の移動による)とそれに伴う二次的神経細胞死と考えられている。これまでの報告や申請者の研究により、AQPが脳浮腫に関与している可能性は示唆されているが、その発生機構は十分に解明されていないのが現状である。本研究では、正常AstにおけるAQPの発現調節機構に関して、AQPの発現変化に対する細胞内情報伝達系の関与に注目し、基礎的データを収集することを目標とした。さらには、病的状態(脳浮腫)におけるAQPの発現変化と細胞内情報伝達系との関係を詳細に検討することにより、AQP発現の調節を主眼においた新しい脳浮腫治療法確立を目指した。
平成17年度は、(1)各種阻害薬によるAQP発現調節、(2)AQP阻害薬の検索、以上2点について研究を実施する。
(1)各種阻害薬によるAQP発現調節
平成16年度の研究により、AQPの発現調節は、転写因子であるNF-κBによって行われている可能性が示唆された。さらに本年度の研究により、培養Astに各種NF-κB阻害薬を投与したところ、AQP発現は低下することが明らかとなった。
(2)AQP阻害薬の検索
さらに、培養Astに低酸素を負荷する細胞障害系にNF-κB阻害薬を添加したところ、細胞障害が軽減することを観察した。今後、脳虚血モデル動物にNF-κB阻害薬を投与し、脳障害が軽減することを確認する必要がある。
