Research Abstract

TGF-βは受容する細胞によって応答性が異なる。一般に上皮系の細胞には細胞増殖抑制を惹起し、間葉系細胞には増殖促進を誘導する。また癌環境においては、初期では癌の増殖を抑制し、進展した環境では悪性度を憎悪するように働くことが近年、明らかになってきている。この1つの機序として癌細胞が浸潤する際、間葉系様の細胞に変化(EMT : Epithelial mesenchymal transition)し、転移した組織では再び基の細胞様に戻る(MET)機構が知られているが、TGF-βがこのEMTを積極的に引き起こすことが知られるようになった。そこで、上皮細胞であるNMuMG細胞を用いて検討すると、TGF-βによってEMTが誘導され、TGF-βの低分子抑制物質で、この誘導を解除できた。さらにこの分子メカニズムを検討すると、HLH型転写因子因子が非所に重要であることが判明した。しかしながら詳細な経時的な検討を行うと、HLH転写因子よりも他のzinc finger型転写因子が上皮マーカーであるE-cadherinの転写調節と非常に関連していることが判明し、その転写因子の過剰発現やノックダウン法により機能解析を行った。また、このzinc finger型転写因子のプロモーターをクローニングし現在TGF-βによる制御領域や制御に関する転写因子の同定を試みている。またEMT誘導時にE-cadherinはプロテアソーム阻害剤によって膜での局在が安定化することが判明し、現在この分子機構を検討すると共に、TGF-βシグナルに関与するE3リガーゼとの関連性を分子生物学的に検討している。
