Research Abstract

腫瘍壊死因子(TNFα)による脳虚血時における神経細胞死誘導の機能的意義は不明の点が多い.本研究は,TNFαの神経細胞死誘導とその細胞死の機構を解明することを目的とし,TNFαおよびレセプターの一過性局所脳虚血(tMCAO)時における局在をタンパク,遺伝子レベルで調べた.またTNFα遺伝子欠損(TNF KO)マウスを用いてtMCAO後の脳梗塞の比較および神経細胞死誘導機構を形態学的,生化学的に明らかにした.さらにIL-1とTNF-αに共通し下流域に存在する細胞接着因子(intercellular adhesion molecule-1;ICAM-1)に関してその分子機構の詳細を検討した.
tMCAO後,TNFαの遺伝子発現の上昇は脳虚血中と再潅流6時間以降の2相性に見られた.その発現細胞は主に神経細胞であり,わずかにマイクログリアおよびオリゴデンドログリアにも観察された.一方,TNF receptor Iは早期では主に神経細胞に発現するが,時間の経過に伴いアストログリアの陽性比率が増加した.TNF KOマウスは有意に虚血性神経細胞死を抑制した.TNFαは虚血時に産生され神経細胞死誘導作用のあることが明らかとなった.TNFα KOはカスパーゼ活性の低下やアポトーシスのマーカーであるTUNEL陽性細胞の減少が認められた.さらにICAM-1は脳虚血後48時間目までは脳梗塞内の血管内皮細胞に発現されるが,48時間以降,血管に加えアストログリアに強く発現した.血管性ICAM-1はIL-1RIの発現と一致し,虚血後期発現アストロサイトのICAM-1はTNFRIと共存し,それぞれ異なる時期に異なる細胞で制御していることを示した.
