Research Abstract

我々は、鳥類のウズラの脳からRFamide抗体免疫陽性物質の単離・同定を試みた結果、12アミノ酸残基からなる新規ペプチドを同定することに成功した。詳しく解析したところ、このペプチドは正中隆起外層に終末する視床下部室傍核ニューロンに局在しており、生殖腺刺激ホルモンの放出を抑制することが分かった。これは脊椎動物から初めて見出されたものであり、生殖腺刺激ホルモン放出抑制ホルモン(Gonadotropin-inhibitory hormone ; GnIH)と名付けた。in vivoとin vitroの最近の解析から、GnIHは、脳下垂体細胞に直接作用して生殖腺刺激ホルモンの放出を抑制する経路と、生殖腺刺激ホルモン放出ホルモン(GnRH)の作用を抑制することによる間接作用の2つの作用経路の存在が示唆された。GnIHの作用機序を明らかにするためには、GnIHレセプターの同定が必要不可欠である。
本研究では、哺乳類のレセプター遺伝子の情報をもとに、鳥類のウズラにおけるGnIHのレセプターを同定することを試みた。哺乳類GnIH同族ペプチドのレセプタータンパク質のアミノ酸配列に基づき、縮重プライマーを作製し、視床下部組織を用いて、GnIHレセプターのcDNAの単離を行った。その結果、GnIHレセプターと思われる7回膜貫通型レセプターのクローニングに成功した。次に、クローニングされたレセプターがGnIHのレセプターであるかを確認するために、クローニング出来たレセプターを培養細胞に強制発現させてレセプター結合実験を行い結合親和性を解析した。その結果、GnIHレセプターであることが確認された。このGnIHレセプターmRNAは、脳下垂体と視床下部領域に発現していた。これらの結果から、GnIHの脳下垂体への直接作用と視床下部への間接作用が初めて示された。
