Research Abstract

天然および非天然の生物活性物質には複雑な環骨格を有する化合物が多い.複雑な骨格を有する含窒素複素環,中でも縮環型化合物や中員環化合物を自在に合成できる手法を開発することは,創薬を指向した基礎的合成研究として極めて意義深い.報告者は,これまで合成困難であった複雑な複素環・中員環を基本骨格とする生物活性物質を合成するための新たな環化反応の開発を目的として研究を行い,平成16年度に以下の成果を得た.
(1)パラジウム触媒を用いたエンアレンの新規閉環反応の開発
報告者が開発した還元的アレン合成法を用いて,種々のエンアレンを合成し,パラジウムによる閉環反応を検討した.その結果,アレンとオレフィン間で炭素-炭素結合が生成し,さらにベンゼン環や複素環上への連続的な閉環が進行し,多環式ベンゾイソインドール誘導体が一挙に得られることを見出した(Chem.Eur.J.,in press).さらに,反応条件を変えるだけで同一のエンアレンからピロリジン体とシクロプロパンを作り分けられることをを明らかにした(J.Org.Chem.,2004,69,4541-4544).
(2)ハロアレンを用いた含窒素中員環の新規合成法の開発
報告者は,ブロモアレンがパラジウム触媒とアルコールの存在下,アリルジカチオン等価体として機能しうることを見出した.この新しい反応性を利用して,分子内に求核部位を有する種々のブロモアレン誘導体の反応を検討した結果,様々なヘテロ中員環を合成することに成功した(J.Am.Chem.Soc.2004,126,8744).さらに,スルファミドを有するブロモアレンを用いたタンデム型閉環反応を検討し,二環性スルファミドの一挙構築にも成功した(Angew.Chem.Int.Ed.2005,44,1513).
