Research Abstract

脈絡叢は脳脊髄液を産生する器官であるが,その中枢神経系における役割の解明は進んでいない。脈絡叢の特性を調べる目的で行ったサブトラクション法,RT-PCRにより,脈絡叢がアミロイドベータ凝集抑制因子,脂質代謝関連因子,細胞移動促進因子,解毒代謝因子,神経栄養因子を豊富に発現していることを明らかにした。脈絡叢がこれらの機能分子を産生し中枢神経環境を適正化しながら中枢神経機能維持を担っていることが示唆される。
今回我々は,生体内における脈絡叢の中枢神経に対する機能をin vitroで評価するため,脈絡叢上衣細胞とニューロンまたは神経幹細胞との非接触性共培養を試みた。これにより脈絡叢上衣細胞が(1)ニューロンのa)突起伸長を促進,b)突起数及びその分枝数を増加,c)神経細胞死を抑制すること,(2)神経幹細胞の分化誘導時にa)細胞死を抑制,b)ニューロンへの分化を支持することを明らかとした。更に,脈絡叢上衣細胞の培養上清中にニューロンの突起伸長,細胞死抑制を促進する因子を有することを証明した。
一方,ラット脳虚血モデルに対し培養脈絡叢上衣細胞を経脳脊髄液的に移殖することにより,梗塞巣の進展を抑え,行動学的な回復が得られることを明らかとした。移殖細胞は脳室内に留まり,損傷部には認められないにもかかわらず,アポトーシス細胞や炎症細胞の出現を抑制していた。更に,脳実質において,cREB,bcl-2の発現を誘導し,IL-1β,TNF-α,iNosを抑制することがわかり,移殖細胞から分泌される様々な液性因子が,脳脊髄液を介して脳虚血損傷に伴うアポトーシス,炎症,酸化ストレスを複合的に抑え,神経保護機能を発揮したものと考えられた。
今後,中枢神経機構維持能力を持つ脈絡叢上衣細胞の移殖により,様々な中枢神経疾患における神経保護と内在性神経幹細胞賦活化による再生を効率的に図る道が拓かれるものと期待している。
