末梢神経損傷によるtPA発現と細胞外基質蛋白の改変及び疼痛発生への影響

Research Abstract

現在までに明らかにしたことは以下の通りである。
1.末梢神経損傷後に分泌型のセリンプロテアーゼであるtissue type plasminogen activator (tPA), urokinase plasminogen activator (uPA) mRNAが後根神経節の主に疼痛受容を担うタイプのニューロンにおいて発現誘導されることがわかった。uPAの発現は末梢神経損傷後24時間以内に有意に発現誘導され、損傷後3日をピークとするのに対しtPAの発現は14日を越えて長期に持続することがわかった。また、tPAを発現しているニューロンは再生関連分子であるgrowth associate protein 43 (GAP-43)と高率に共存を示し、末梢神経再生を含んだ形態変化に関与している可能性を見いだした。
2.後根神経節で発現誘導されたtPA,uPAのうち中枢へ運ばれるのはtPAであることをin situ zymographyにより明らかにした。tPAの免疫陽性は脊髄後角I-II層のプレシナプスに検出され、酵素活性の存在がシナプス分泌による可能性をしめした。
3.tPAの酵素活性を特異的に抑制する薬剤であるtPA-STOPの髄腔内慢性投与により、末梢神経損傷後の脊髄後角ニューロンの興奮性の増加を抑制し、また同様に慢性投与したtPAの酵素活性阻害剤は末梢神経部分損傷モデルラットの疼痛行動を有意に抑制するが、発症初期(〜12日)に限定した濃度異存的な効果であった。
