劇症型溶レン菌感染症起因菌が産生するホスホリパーゼA2の病原因子としての機能

Research Abstract

昨年度までの研究から、Streptococcus pyogenesのゲノム上にコードされているフォスフォリパーゼ様のタンパク質PLA_2(Sla)が、マウスの筋肉細胞に対して細胞障害活性があることを確認した。Streptococcus pyogenes由来のPLA_2をコードするsla遺伝子の突然変異株を用いて、どの組織由来の細胞に作用するかを調べるため、ヒト由来の様々な培養細胞(肺上皮細胞(A549)、横紋筋細胞(A673)、喉頭上皮細胞(CCL-138)、肝臓上皮細胞(HepG2))を用い、細胞特異性を調べたが、野性株あるいは突然変異株を各細胞へ感染させたとき明らかな変化は見られなかった。本年度の研究では、sla遺伝子産物が分泌される培養条件およびsla遺伝子産物の最適pHなどの活性化条件の検討を行った。活性化の条件は、細胞が破壊した場合に培養液中に流出する細胞内タンパク質であるLactate dehydrogenaseの活性を指標として測定した。最適pHを調べるため、培地をpH6.5〜8.5に調整し、増殖した培養細胞の培地交換をし、酵素を添加した。この酵素を培養細胞へ添加した結果、pH7.0とpH8.5でLactate dehydrogenaseの活性が上昇した。しかしながら、pH6.5,pH7.5,pH8.0では、Lactate dehydrogenaseの活性が減少した。これらの結果から、この酵素は、pH7.0とpH8.5で細胞障害活性をもつことが示唆された。
