Research Abstract

本研究では,in vivo計測の利点を生かし,腎疾患発症前の段階からラジカル消去能を各臓器毎にESR画像化解析を行い,リアルタイムの酸化ストレス変化を追跡し,病態発症・進展への関与を明らかにすることを目的とした.
1.ストレプトゾトシン(STZ)投与Nrf2遺伝子欠損マウスにおける検討
In vivo ESR測定の結果STZ投与Nrf2遺伝子欠損マウスではSTZ投与野生型に比べスピンプローブCarbamoyl-PROXYLの消失半減期がSTZ投与1週後では短縮し,2週以降は延長に転じた.STZ投与モデルはNOの強い関与が知られているが,STZ投与Nrf2遺伝子欠損マウスにNO合成酵素阻害剤L-NAMEおよびマンニトールを投与するとCarbamoyl-PROXYL消失半減期は回復した.このことよりSTZ投与Nrf2遺伝子欠損マウスではペルオキシナイトライト-ヒドロキシラジカル系を介した酸化ストレス亢進が存在することが示唆された.
2.虚血再潅流障害による急性腎不全モデルにおける検討
片腎摘出,片腎30-45分の虚血再潅流モデル作成し,ESRイメージングにより腎局所での酸化ストレス変動評価の可能性を検討した.Carbamoyl-PROXYL静注後約30分間に4-6回程度のESRイメージが撮像可能であり,静注直後の像から特定された腎,肝など特定領域の画素値を経時的に片対数グラフにプロットするとR^2=0.90程度の相関関数を持って近似可能であった.この方法により得られた腎の半減期は,虚血再潅流後3日目に延長し,7日目では部分的な回復に留まることを示した.一方従来のL-band ESR測定では上腹部でのCarbamoyl-PROXYL半減期は7日目には虚血前のレベルに戻り,腎でのESRイメージングの結果と解離した.このため臓器ホモジネートを作成し,フェリシアン化カリウムを用いて臓器中の還元能をex vivoで測定したところ,その結果はESRイメージングの結果と合致するものであった.以上よりESRイメージング法が従来のL-band ESR法よりも腎障害時の酸化ストレス評価として適切であると考えられた.
