Research Abstract

昨年に引き続き、我々がこれまでに樹立したヒト消化器癌転移細胞バンクから、肝転移あるいは腹膜播種能の最も強いHPC-4H4(高肝転移株)とHPC-4P4a(高腹膜播種株)を用い、この2種類の高転移細胞株からそれぞれmRNAを抽出し、cDNAを合成し、これを哺乳類細胞発現ベクターの中に組み込み、それぞれのcDNA libraryを作製した.これらを共通の親株である低転移性の細胞株HPC-4に遺伝子導入し、強制発現させた.導入効率の高いものから順に、肝転移株・腹膜播種株ごとそれぞれ10種類を目標に、ヌードマウスの脾臓内あるいは腹腔内に接種した.
現在までに一度の接種で、高転移株と同等の転移率を示した細胞集団は存在していない.しかし高転移細胞株ほどではないが、転移を認めた集団も数種得られ、これをin vitroの系に戻し、継代培養し、細胞株として樹立した.またlibralyよりさらに多数を高転移株に遺伝子導入し、強制発現させた細胞集団の作製を試みている.
また癌腫を変え、胃癌細胞株を用いて、同様の工程を行うことも検討中である.
