Research Abstract

in vitroならびin vivoヌードマウスモデルにてTS-1と放射線の併用療法における抗腫瘍効果発現のメカニズムを解析し、さらにTS-1と放射線の併用が生存シグナルであるp-Aktおよび、DNA二重鎖切断修復関連因子Rad50、Ku70、Ku86、DNA-PKcsへ与える影響につき検討を行った。
[方法]in vitroにおいてヒト口腔扁平上皮癌細胞(B88)をTS-1(10μg/ml)と放射線(15Gy)を単独または併用にて処理し、増殖能(MTT法)、コロニー形成能(clonogenic assay)さらにアポトーシス誘導能(Hoechst33258染色、colorimetric assay)に与える影響について検索し、さらにB88をヌードマウス背部皮下に移植した後、TS-1(10mg/kg/day,3週間連日経口投与)と放射線(1.5Gy/day,5times/week,3週間照射)を単独または併用にて治療し、腫瘍体積ならびに体重の変化を測定するとともに、残存腫瘍におけるp-Akt、Rad50、Ku70、Ku86、DNA-PKcsの発現を免疫組織学的に検索し、アポトーシスの誘導はTUNEL法により検索した。
[結果]in vitroにおいてTS-1と放射線との併用処理にて、著明な増殖及び分裂死の抑制、さらにcaspase-3,-8,-9の活性化を介したアポトーシスの増強が認められた。また、in vivoにおいてTS-1と放射線、との併用療法にて有意な抗腫瘍効果が見られ、この際、免疫組織染色法にて残存腫瘍におけるp-Akt、Rad50、Ku70、Ku86、DNA-PKcsの発現が減弱し、さらにTUNEL法にてDNA断裂した腫瘍細胞が有意に増加していた。なおTS-1と放射線との併用は、TS-1先行後放射線照射を行った方がより効果的であった。
[総括]以上より、TS-1とIRの併用効果は、P-Aktを介した生存シグナルの抑制、さらにはRad50ならびにDNA-PKcs複合体を介したDNA二重鎖切断修復機構の抑制が関与し、その結果、放射線増感効果を高め、アポトーシスを増強し、抗腫瘍効果を発現している可能性が示唆された。
