Research Abstract

ミクロ制御として、表面構造が1〜10μmの幅と0.5〜1.5μmの深さを持つmicrogrooveを付与した培養基質上で骨性細胞を培養し、電子顕微鏡、共焦点レーザー顕微鏡による形態学的観察を行った結果、grooveに沿って細胞を伸展させ、さらには細胞が産生した石灰化基質の沈着もgrooveに沿って沈着させられ、幅が1、2μm上での細胞はridge上に強く接着していることが証明された。in vitroでの三次元的な骨組織構築を目的として、この細胞動態を利用して二重構造(台形groove:幅100μm+v字形microgroove:幅2μm)を有する基質(multi-groove)を作成し、in vitroにおける骨様組織の三次元的形態付与を実現させた。ナノ制御として、培養基質にプラズマ表面改質装置を用いた後にBMPあるいはfibronectin含有PBSを2時間留置した。その結果、蛍光抗体法を用いた方法によりBMPあるいはfibronectinが基質に固定されていることが確認され、Q-senseを用いた物理学的な方法により44.2ng/cm^2量のBMP、260.0ng/cm^2量が固定されていることが証明された。BMP固定培養基質上で、骨性細胞の培養実験を行い、有意に骨芽細胞の機能発現の指標となるALPの活性が上昇し、骨関連タンパクであるオステオカルシンのmRNAの発現も増加した。Fibronectin固定培養基質上での細胞接着に関しては、初期における接着細胞数が有意に多い結果となった。
