Research Abstract

失った歯牙関連組織を再生・復元させることは歯科界にとっての重要なテーマであり目標である。本研究では、歯牙周囲組織再生の細胞源として、不要なった智歯、歯牙などから採取可能な歯髄組織に着目した。この歯髄組織中には歯髄幹細胞(DPSCs)が存在し、未分化間葉系幹細胞(MSCs)と類似した特性を持つとされ、象牙質、神経、脂肪などに分化する多様性も指摘されている。まず、このDPSCsの表面抗体を検討し、MSCsと同様な表面抗体の発現を認めた。また、骨、脂肪などに分化誘導を行った結果、それぞれの細胞に分化することも確認された。さらに、同一患者から採取した口腔粘膜(hOMCs)、hDPSCs、hMSCsにおける分化誘導における違いを各種組織再生についての可能性を遺伝子発現、機能的な面からも検討した。また、硬組織、象牙質特異的遺伝子発現を定量的RT-PCR法を用いて比較した結果、DPSCsはMSCsより象牙質特異的遺伝子(DSPP、DMP-1など)の遺伝子発現が培養後期(24日後)において高値を示した。そこで、これら細胞のうち、特に歯牙周囲組織への再生に応用可能と考えられるhDPSCs、hMSCsの特性の違いについて、遺伝子プロファイルとクラスタリング解析により検討した。この結果、MSCsに比べ、DPSCsは発現に変化が見られた遺伝子群にはcollagen, MMP, MSX1, BMP2,4, PAX9, BGPなどが認められ、未分化なMSCsに比べ、硬組織へ分化誘導したDPSCsに特徴的であるclusterにおけるクラスタリング解析においても、cell signaling, communication関連遺伝子TNFSF11, WNT5a, metabolism関連遺伝子PTGS2, SYKや他にPAX9, TBX2に関する遺伝子群の発現が認められた。さらにDPSCsを吸収性セラミックスなどと動物の背部皮下に移植すると象牙質様硬組織の形成が確認された。このような結果から、細胞工学、遺伝子工学的に骨、象牙質、歯などをDPSCsおよびMSCsを応用することによって、再生できる可能性が示唆された。今後はより詳細に細胞特性を検討すると同時に、目的とする組織再生を効率的に行えるよう、また歯牙硬組織再生を一日も早く実現するように進めたい。
