Outline of Annual Research Achievements

メダカは、飼育の容易さと飼育環境の温度域も広く、海水耐性も強いことから環境毒性試験に広く用いられている。本研究では、放射線、紫外線、内分泌撹乱物質、重金属による刺激後のメダカから血液を採取して血球におけるDNAダメージと血清内での分子変化の解析から生物影響を解明し、突然変異体と野生型を比較することで関連する遺伝子機能を明らかにすることを目指した。
本年度は、成魚の紫外線UVA照射による急性生物影響を赤血球におけるDNA損傷をγ-H2AX とコメットアッセイにより検出した。また、内部組織においても卵巣中の卵形成に急性障害が見られ、眼の網膜、脳、脊髄にも細胞死などの影響もでること、p53変異体では、電離放射線と同様に影響の程度が小さいことを確認した。今後、作製したサンプルからUVA照射した成魚の肝臓、腎臓、脾臓などの組織変化を調べるとともに、特に魚類の腎臓、脾臓、肝臓などに存在する褐色の色素を持ったマクロファージの集塊であるメラノマクロファージセンター(異物や老化した細胞を非特異的に貪食、消化して他の細胞に受け渡す機能を有すると考えられている)のUVA照射後の形成についても検討している。
また、4-Nonylphenol (4-NP)にばく露したメダカ成魚赤血球の形態異常、小核形成とアクリジンオレンジ染色による細胞死の検出を行い、濃度依存的にそれらが増加することを定量化した。先行研究で行なったナマズへの影響と比較したところ、メダカはより4-Nonylphenol (4-NP)に対する感受性が高く、環境中の毒性化合物の検出系として適していることが示された。
