Outline of Annual Research Achievements

がんのドライバー変異に関し、タンパク質アミノ酸変化を起こす変異と共に、調節領域上の変異を研究する必要がある。本研究では、まず、人の様々ながんで、2,000を超える因子のモチーフマトリックスを検索し、ヒトゲノム全体の上で、各転写因子の結合のしやすさを計算することで、転写因子結合部位の変異に伴う機能アノテーションのリストを作製した。続き、COSMICリポジトリを検索し、プロモーター領域上にある、アノテーション付き体細胞突然変異を網羅的に取得した。そして、上記の結合部位に体細胞突然変異を当てることによって、がん種ごとに、変異の蓄積の多い転写因子結合部位を調べた。その結果、プロモーター領域内の体細胞突然変異とゲノムとの相互作用が、そしてがんごとに高度に突然変異した因子が認められた。さらに、がんの発症に関与する翻訳後修飾マーク「サクシニル化」を調べた。このために、サクシニル化リジンを含むというアノテーション付きのタンパク質のリストを検索し、各タンパク質について、アミノ酸の構造的および進化的特徴を計算した。その特性の計算には、アクセス可能な表面積、ねじれ角、二次構造、および位置特有のスコアリング行列を考慮した。各リシン残基は、その15個の下流のアミノ酸および15個の上流のアミノ酸を考慮したベクターとして表現される。このようにして、サクシニル化および非サクシニル化リジンの大きなデータセットを得て、サクシニル化部位を正確に予測することができる新しい計算法を実装し、論文化することができた。これらから得られた一連の結果を、Analytical Biochemistry、Journal of Theoretical Biology、BMC Genomicsなどの科学論文誌に掲載することができた。
