Outline of Annual Research Achievements

カルパインはほぼ全ての真核生物と一部の原核生物に存在する細胞内調節性プロテアーゼのスーパーファミリーである。良く保存された触媒ドメインの両端に多様な機能領域を保持し、ヒトでは15遺伝子から6構造型の分子が発現する。その構造多様性により、同一ファミリー内においても各分子が際だった特徴（超短寿命、非プロテアーゼ機能、分子間相補能等）を示すという、他のプロテアーゼには見られない特質がある。近年、遺伝学、分子生物学の発展が、各分子種の特徴と機能の関係解明を強力に促進した結果、カルパインのバイオロジーは新たなフェーズに突入した。そこで本研究では、申請者ら自身が明らかにしてきたカルパイン各分子種の生理機能をより精緻に系統化し、得られた知見の生物情報学的な解析からカルパインが生物の特性にどう関わるのかについて理解することを目指している。
生物学的な意義という観点から、CAPN3/6と骨格筋、CAPN8/9/12と胃腸/皮膚、CAPN7と肝臓/脾臓、CAPN11と精巣/精子の組み合わせに重点を置き、各カルパイン遺伝子改変マウスの表現型について、プロテオミクスを基軸とした生化学的解析を行い、カルパインの作用点に関わる分子群を解析している。従来型カルパインについては、酵素学および生物情報学を組み合わせて、網羅的に基質特異性を解析している。これらの継続的な解析の結果、カルパインの基質切断予測を含め、カルパインの基質認識機構の一部を明らかにすることができ、現在二報目の論文を作成中である。今後は、これらの解析結果をもとに、カルパインのドメイン構造の共通性と独自性を検討し、構造-機能相関の観点から各ドメインの生理機能を解析していく。そして最終的には、分子間での機能と作用機序の共通点を検討し、生物界におけるカルパインスーパーファミリーの存在意義を考察する予定である。

Strategy for Future Research Activity

計画は概ね順調に進展しているので、今後も計画に従って、力の限り研究を遂行していきたい。具体的には、下記の通りである。既述の通り、カルパインはほぼ全ての真核生物と一部の原核生物に存在する細胞内調節性プロテアーゼのスーパーファミリーである。保存された触媒ドメインの両端に多様な機能領域を保持し、その構造多様性により各分子が様々な興味深い特徴（超短寿命、非プロテアーゼ機能、分子間相補能等）を示すという、特質がある。そこで本研究では、カルパイン各分子種の生理機能をより精緻に系統化し、得られた知見の生物情報学的な解析からカルパインが生物の特性にどう関わるのかについて理解することを目指す。即ち、生物学的意義から、特定のカルパインと特定の組織との組み合わせに重点を置き、各カルパイン遺伝子改変マウスの表現型について、プロテオミクスを基軸とした生化学的解析を行い、カルパインの作用点に関わる分子群を解析していく。従来型カルパインについては、酵素学および生物情報学を組み合わせて、網羅的に基質特異性を解析していく。そして、これらの解析結果を総合し、カルパインのドメイン構造の共通性と独自性を検討し、構造-機能相関の観点から各ドメインの生理機能の解明を目指す。最終的には、分子間での機能と作用機序の共通点を検討し、生物界におけるカルパインスーパーファミリーの存在意義を考察する予定である。
