Outline of Annual Research Achievements

昨年度は、メタフェタミンの長期使用による退薬症状におけるマウス側坐核のBDNF-TrkBシグナルの役割についての研究成果を論文として発表した。今年度では、引き続き覚せい剤精神病のモデル動物を用いて、BDNF-TrkBシグナルや脂質代謝に関わる可溶性エポキシド加水分解酵素の役割を調べた。
覚せい剤の長期使用に伴う精神神経障害及びうつ症状などの離脱症状に対する根本的な治療薬がないのは現状である。昨年度の研究成果より、覚せい剤の繰り返し投与による離脱症状には、側坐核のBDNF-TrkBシグナルの亢進が関わっている可能性を示唆された。今年度は、iTRAQ又は次世代シークエンサーの解析より、覚せい剤の繰り返し投与によって側坐核で変動する幾つかのタンパクや遺伝子を見つけた。さらに、覚せい剤の繰り返し投与による行動異常には、脂質代謝に関わる可溶性エポキシド加水分解酵素が関わっている可能性を見出した。また、可溶性エポキシド加水分解酵素阻害剤の投与は、覚せい剤の長期投与で引き起こす行動異常を改善することを見出した。
現在、可溶性エポキシド加水分解酵素の遺伝子欠損マウスを用い、覚せい剤精神病における可溶性エポキシド加水分解酵素の役割とその分子メカニズムの解明を進めている。予想通り順調に進んでおり、来年度に研究成果をまとめ、論文投稿する予定です。
