交尾刺激による生殖幹細胞の増加を司るステロイドホルモンを介した神経機構の追究

Outline of Annual Research Achievements

卵が安定的に供給されるためには、卵巣に存在する生殖幹細胞の分裂と維持の制御が鍵である。一方で、生殖幹細胞の分裂が、個体を取り巻く外的環境要因によってどのように制御されるのかは不明な点が多く残されている。申請者は、キイロショウジョウバエ成虫メスにおいて、生殖幹細胞の増殖が交尾によって引き起こされることを明らかにした。交尾依存的な生殖幹細胞の増殖は、オスの精液中のペプチド成分によって引き起こされる「性ペプチドシグナル経路」およびメス卵巣での「ステロイドホルモン生合成」によって制御されることを証明した(Ameku and Niwa, PLoS Genet. 2016; Ameku et al., Fly 2017)。

交尾依存的な生殖幹細胞の増殖を制御する因子を探索するため、284の膜受容体遺伝子を標的としたin vivo RNAiスクリーニングを実施した。その結果、機能阻害により交尾後の生殖幹細胞の増殖が抑制される遺伝子群を得た。これらの遺伝子群には、これまで生殖幹細胞の維持に重要な「ニッチシグナル」として機能する遺伝子が含まれていた一方、生殖幹細胞の制御への役割が未知の遺伝子が含まれていた。その中でも腸内分泌細胞から放出されるホルモンに着目し、詳細な機能解析を行った。その結果、交尾後の生殖幹細胞増殖を制御する組織間コミュニケーションのメカニズムの一端を明らかにした(Ameku et al., 査読中)。これらの結果は、従来までの分子レベルでの生殖幹細胞の研究分野に、新たに行動・神経・生理レベルでの観点を与える意義があると考えられる。
