Outline of Annual Research Achievements

再生医療の分野では、iPS細胞などの多様性幹細胞から目的の臓器細胞へと分化誘導を行う試みが行われている。しかしながら、特定の臓器細胞へ分化させた細胞群に未分化な細胞が残存することによって、体内で腫瘍を形成することが危惧されている。すなわち、必要な、あるいは不要な細胞を分離する技術が細胞移植や再生医療の分野において強く求められている。
そこで、本研究では、所属研究室で開発した電気化学的原理に基づいた電位印加によって培養表面から細胞を素早く脱離する技術を応用し、選択的な細胞分離法の確立に取り組んだ。これまで、電気化学細胞脱離に用いるオリゴペプチドの片末端には、多くの細胞が認識できる細胞接着アミノ酸配列(RGD)を配置していたが、本研究ではこれに代わり、アプタマーを配置した。これによって、細胞集団の中から特定の細胞を表面に接着させるのである。ここでは、モデル細胞として肝癌細胞と特異的に結合するアプタマーを用いた。この基板上に肝癌細胞と線維芽細胞を同数播種し、接着したそれぞれの細胞数をカウントし、細胞選択性を評価し結果、85%の選択率で肝癌細胞を選択的に分離できることを示した。また同様に肝癌細胞と正常肝細胞を基板上に播種した結果、98%と高い選択率で分離できることがわかった。さらに、肝癌細胞を金基板に選択的に接着させた後、-1.0 V (vs. Ag/AgCl)を5分間印加し自己組織化単分子膜を脱離させることで、接着した細胞を脱離・回収することができた。さらに回収した細胞の機能評価を行ったところ、通常培養で用いられる酵素処理による回収法と比べて、同等の生存率・機能を示すことを確認した。
