Outline of Annual Research Achievements

B型肝炎ウイルス (HBV) 感染分子機構のうち、特にウイルス感染に必須な宿主との相互作用の理解は充分ではない。申請者はこれまでに簡便にHBV感染を評価出来る新規細胞培養系としてHepG2-hNTCP-C4細胞を樹立した。本研究ではこの細胞培養系を用いて、HBV感染に関わる宿主因子を同定するために、膜タンパク質であるG-Protein Coupled Receptor (GPCR) を標的としたsiRNAスクリーニングを実施した。標的遺伝子の発現抑制に伴いHBV感染が減少したヒット遺伝子のうち、最も再現性良くHBV感染を減少させたものとしてG Protein-Coupled Receptor 160 (GPR160) を得た。GPR160ノックダウン細胞では、HBVと同じく胆汁酸トランスポーターであるsodium taurocholate cotransporting polypeptide (NTCP) 依存的に細胞内へ侵入するD型肝炎ウイルスの感染も低下していた。この細胞ではNTCP依存的なHBV エンベロープの細胞表面への吸着も低下していた。また、GPR160ノックダウンによりNTCPのmRNA量を減少させる事無く、細胞膜上のNTCPタンパク質量が減少してした。加えてNTCP本来の機能である細胞への胆汁酸の取り込み量も低下した。これらの結果は、GPR160がNTCP の細胞膜上発現を制御する事でHBV感染に寄与する宿主因子であることを示唆する。
上記研究により、感染に必要なウイルス-宿主間相互作用の理解のみならず、NTCPおよびこれが関わる胆汁酸取り込みの制御機構の一端が明らかになると期待される。
