Outline of Annual Research Achievements

本研究では,基板表面に細胞の接着領域をパターニングすることにより神経細胞の配置と軸索の伸長方向を制御し,脳の機能を発現する最小パターン神経細胞ネットワークを構築することを目的としている.脳内の神経細胞の電気的特性において,個々の細胞は異なる周波数特性を持ち特定の周波数信号により強く応答することで信号を効率よく処理している。本年度は軸索方向を制御したパターンを用いて神経細胞の形状を制御し,神経細胞の膜インピーダンスの強度変化を試た.昨年度と同様に軸索と樹状突起の極性制御パターン上に培養した神経細胞と,パターンを施していない基板上に培養した神経細胞の膜インピーダンスとを比較した。神経細胞の膜インピーダンスの周波数特性を調べるために,刺入した電極から周波数が0.1 Hzから100 Hzに変化するChirp電流信号を入力し,その時の膜電位の変化を測定した。これらの入出力信号をフーリエ変換することで膜インピーダンスの周波数特性を計算した。培養7日目ではパターン神経細胞と非パターン神経細胞ともにlow-pass特性を示し,膜インピーダンスは同程度であった。より形状に差が出た培養16日目ではパターン神経細胞が10-40 Hzの低周波数帯において,高いインピーダンス示した。シミュレーション結果においてもパッチクランプ実験で得られた結果と同様にlow-pass特性が見られた。実験結果と計算結果において膜インピーダンスはlow-pass特性を示したが,形状の制限によるインピーダンス強度の変化は大きく異なった。以上より,神経細胞の電気的素子特性である膜インピーダンスを制御できるようになった。この技術により機能的な神経活動を有する最小の神経回路の構築が現実的となった.
