Outline of Annual Research Achievements

大脳皮質の神経回路は生後初期に神経入力依存的に大きく再編されることで高度な情報処理能力を獲得する。しかし、生体内でどのような過程を経て再編が達成されているのかはほとんど不明であった。その実態解明のため、私はマウス大脳皮質体性感覚野の「バレル」構造をモデルに研究を進めてきた。個々のバレルでは、1本のヒゲからの情報を伝達する視床皮質軸索がクラスターを形成しており、その周りを取り囲む神経細胞が単一のクラスターに向けて樹状突起を伸ばすことで、1本のヒゲ情報が選択的に処理されている。バレルは生後1週間の内に神経入力依存的に形成され、その形態から回路の機能成熟を検出できるため、発達期の入力依存的回路再編機構を理解するための有用なモデルとされてきた。
本研究課題では、バレル形成過程を生きた個体脳内で直接観察するという独自のアプローチを進めてきた。これまでに、生後2,3日齢から生後6日齢までというバレル形成が劇的に進む期間をターゲットとした新生仔大脳皮質の長期生体イメージング手法を確立し、神経細胞が1つのバレル選択的に樹状突起を展開するようになる過程での動態の定性/定量解析を行ってきた。本年度はさらに、ヒゲからの情報を脳に伝える眼窩下神経を外科的に切断した新生仔マウスの長期生体イメージング、蛍光カルシウムプローブを用いた神経活動の生体イメージングとその解析等を行った。新生仔の大脳皮質で入力依存的に神経回路が再編される過程を1匹の個体で通して直接観察し、その機構の一端を解明したこれまでの成果は、論文としてまとめ、現在投稿中である。
