Outline of Annual Research Achievements

平成29年度は, 細胞環境におけるタンパク質機能解析に重要となるレアイベントを再現するために開発した計算手法を適用し, タンパク質機能解析を実施した. タンパク質の機能発現に重要な構造変化は「レアイベント」であり,通常の分子動力学(MD)で追跡可能な時間より長時間の確率過程で観測される.レアイベントを計算機上に再現し, 原子レベルの解像度で解析することは, タンパク質の機能発現を理解する上で非常に有益である. これまでに開発した計算手法は, 生体分子の構造サンプリングに特化した設計であったため, 探索構造の定量的な評価が難しいという課題があった. そこで課題克服に取り組んだ. 具体的には, 構造探索法でサンプルした生体分子構造の構造遷移をマルコフ過程と仮定し, 遷移行列を最尤推定により見積もり定常状態を求めることで自由エネルギー評価を可能にした. この拡張により, 様々な生体機能に関係する構造変化の自由エネルギー地形を解析することが可能となった.
開発手法適用によるり, 細胞分裂ダイナミクスに関係しているタンパク質FtsZの構造変化を解析した. FtsZは, 細胞膜の内側にリング状のフィラメントを形成し, 離合集散を繰り返すことで細胞膜に陥入を生じさせるが, 分子メカニズムには未解明な部分が多い. そこで, 細胞環境ダイナミクスを解析するため, 黄色ブドウ球菌FtsZの結晶構造をもとに開発した計算手法を適用し, FtsZの動的秩序解析を行った. 具体的には, X線結晶構造解析から決定されている2状態(T-state, R-state)を始状態と終状態として, 開発した遷移経路探索法を適用し, FtsZの構造変化メカニズムを解析した. 研究成果は, 論文 (J. Struc. Biol., vol. 198, pages 65-73 (2017)) として出版された.
