Outline of Annual Research Achievements

本研究は、歯根膜組織における間葉系幹細胞もしくは前駆細胞集団の局在や機能を同定し、同細胞群がどのようなプロセスで様々な間葉系細胞へと分化するのかを明らかにすることで、歯周組織の恒常性維持と再生機構の理解を深めることを目的としている。本年度は主に以下の3つの成果を得た。
1.Dermo1-Cre；R26-Tomatoマウスの歯周組織の組織学的解析により、歯根膜組織中の間葉系細胞のほぼすべてがTomato陽性であったことから、「H2BGFP標識保持細胞」の同定に用いるCreマウスをDermo1-Creとした。Dermo1-Cre; R26-tTA;TetO-H2BGFPマウスを交配により作製し、6週齢からドキシサイクリンを投与し、経時的に解析したところ、6ヶ月経過するとほとんどの細胞でGFP標識が消失していることが明らかとなった。
2.歯根膜細胞の初代培養プロトコルを確立し、Nestin-GFPマウス由来の歯根膜細胞の単離およびセルソーターによるGFP陽性細胞の分取・培養、さらにはシングル・セルソーティング法によるNestin-GFP陽性クローンを樹立した。同細胞は高い分化能を持つ間葉系幹細胞である可能性が示唆されており、今後の歯根膜細胞を解析するための有用なリソースとなることが期待される。
3.Nestin-Cre; Nestin-GFP; R26-Tomatoマウスの第二臼歯に5-0絹糸を結紮することにより、歯槽骨吸収を惹起する骨吸収モデル実験を行った。絹糸除去後の歯周組織修復過程におけるNestin陽性細胞の系譜を経時的に解析したところ、平常時は毛細血管近傍にのみ位置するペリサイトであったNestin陽性細胞が、修復時には組織全体に広がって、修復組織の大部分を占めることが明らかとなった。
以上の得られた結果を取りまとめ、国内の学会で発表を行った。
