Outline of Annual Research Achievements

間葉系幹細胞(MSCs)は骨、脂肪、軟骨細胞への分化能を有し、運動器に対する移植細胞として最も着目されている。すでに臨床研究においてMSCs移植治療効果の検討がなされているが、術後のリハビリテーションに関する検討は十分になされていない。本研究では、細胞を用いた再生医療の治療効果を、リハビリテーションによって最大限に引き出すことに着眼し、リハビリテーション介入の一つとして用いられる温熱刺激がMSCsの生存能・硝子軟骨分化能・細胞外基質形成能に及ぼす影響を明らかにすることを目的とし、温熱刺激によるMSCs移植後の関節軟骨再生効果を検証するものである。初年度では、実験で用いるMSCsの確立および温熱刺激の持続的負荷がMSCsに与える影響にかかわる実験を計画した。ラット大腿骨骨髄からMSCsを採取し、骨・脂肪・軟骨への分化誘導を試みた。その結果、骨および脂肪へは誘導されたが、軟骨分化誘導においては、軟骨特有の細胞外基質の産生が乏しく、プロトコルの見直しを行った。再検討した実験条件としては、用いるラットの週齢(7週齢~12週齢)、ペレット培養に用いる細胞数(25万~100万細胞/pellet)、軟骨分化誘導専用キットの使用、播種後のMSCsの選別などを検討した。しかしながら、若干の基質産生量亢進は認められたが、良好な軟骨基質形成にまでは至らなかった。現在、軟骨分化誘導に必須であるTGF-βの購入業者を変更して検討中である。MSCsの軟骨分化誘導に予定よりも時間を要しているため、同時にラット骨軟骨欠損モデルの確立のための予備的実験を、予定を早めて開始した。8週齢のWistar系雄性ラットの左右大腿骨滑車面中央部に直径1㎜のバイオプシーパンチで深さ1㎜の骨軟骨欠損を作成したところ、その良好な欠損作成再現性を確認した。今後これらの予備的実験を基に、温熱刺激介入実験を進める。
