Outline of Annual Research Achievements

光誘起の流体効果で生体関連物質の相転移,特異的結合,細胞の状態制御の原理を開拓し,社会で求められるバイオ分析技術に発展させることが本研究の目的である.これまでに金属ナノ粒子固定化ビーズを分散した水中にレーザー光を照射すると,開始からわずか数秒で沸騰現象が起こる条件があることを見出したが,最終年度はマイクロ流路中での圧力駆動流の下で光集積した金ナノ粒子固定化ビーズを熱源として,光誘起バブルによる微量タンパク質の迅速検査に関する新たな知見を後輩との共同研究で獲得して論文投稿を行った.
また,集光したレーザー光照射による集合現象と溶液中に誘起される流れの相乗効果により,金ナノ粒子表面に修飾した一本鎖DNAと浮遊状態の一本鎖DNAの衝突確率を高めることで,DNAの二重鎖形成プロセスを加速できることを2年目に解明したが,タンパク質や異種DNAの混合条件下でもDNA二重鎖形成の光加速が可能であることも示し,遺伝子検査などへの実用展開における重要な知見も獲得した.これらの知見を活かし,DNAチップへの実装を目指した検討も行った.派生テーマに関して後輩にサンプル調製の方法を伝授し,電極間に光集積したプローブ粒子とターゲットDNAの電気検出の迅速化に関する新たな知見も獲得して特許申請にも貢献した(特願2017-095715).
さらに,高温状態で疎水性を示す有機分子を表面修飾した金ナノ粒子内包デンドリマーの特性に注目し,非共鳴レーザー照射による局所加熱により癌細胞表面への集積化に成功した.集積化後に細胞の生死判定を行ったところ,非共鳴条件下であるにもかかわらず,レーザー照射点近傍の癌細胞だけを選択的に死滅できることも明らかにした.この成果は非従来的な光誘導型ドラッグ・デリバリー・システムの新原理開拓につながり,医薬学の発展に貢献し得るものである(国際特許PCT2017-20883).
