Outline of Annual Research Achievements

平成28年度までに磁気分離能とイメージング機能を有する、Ag@FeCo@Agコア@シェル@シェル型ナノ粒子を創生し、哺乳動物細胞へトランスフェクションすることで、オートファゴソームに粒子を送達し、オートファゴソームの磁気分離に成功した。その際、粒子表面は親水性ポリマーで被覆されていたが、オートファゴソームの分離において粒子表面の機能化は重要ではなかった。今後、様々なオルガネラの分離に利用可能な汎用性の高い磁気分離技術の構築を考慮すると、粒子表面を任意のリガンドで修飾することが必要となる。
平成29年度は粒子表面へ任意のリガンドの修飾を可能にするために、粒子表面へアビジンを修飾した。アビジンが修飾された粒子はビオチン標識された任意のリガンドを粒子表面へ修飾することが可能となり、磁気分離可能なオルガネラの種類が広がることが期待される。本研究では最終的に粒子表面へトランスフェリンを修飾した。トランスフェリンは細胞内へ鉄イオンを運搬する役割を担うタンパク質である。細胞膜上のトランスフェリン受容体に鉄イオンを保持するトランスフェリンが結合すると、細胞内の異なるオルガネラを経由して最終的に細胞外へ放出される。そこで、トランスフェリン修飾粒子を細胞へエンドサイトーシスにより取り込ませ、標的となるオルガネラへ粒子が送達されるかを調べた。その結果、トランスフェリン修飾ナノ粒子の流体力学的粒径が約170 nmと大きいため、エンドサイトーシスにより効率的に細胞内へ取り込まれなかった可能性が示された。今後の展望として、タンパク質修飾後も流体力学的流径が小さな粒子を作製する為に、粒子表面の配位子やタンパク質修飾過程を改善することが必要となるであろう。
