Outline of Annual Research Achievements

本研究では、ホルモン分泌を担う下垂体前葉の機能調節機構を幹細胞という観点から解明することを目的に、下垂体幹細胞ニッチ(微小環境)の制御機構解析を目指す。具体的には 1.幹細胞ニッチにおけるephrin-B2/Eph Receptorの解析、2.幹細胞ニッチの単離と制御因子の同定、を行う。本年度の研究成果は以下の通りである。

1.下垂体幹細胞におけるephrin-B2/Eph Receptorの解析: これまでに、ephrin-B2が下垂体の幹細胞ニッチに発現していることに注目し、パートナー分子の同定に成功した。本年度は研究結果を原著論文にまとめ、Cell and Tissue Res.に受理となった。
2.下垂体幹細胞ニッチの単離と制御分子の同定：下垂体幹細胞ニッチの存在割合は全下垂体前葉細胞の1% 以下と非常に低い。そのため，ニッチの性質、ならびにそこで機能する分子を同定するために、ニッチの単離方法の確立が必須である。前年度までに，ニッチの単離方法、ならびに、それら幹細胞ニッチ由来幹・前駆細胞を用いたホルモン産生細胞へのin vitro分化誘導に成功している。本年度は、胚葉を超えた分化能を有するかに関して解析を行った。結果、2次元培養を行うことにより、外胚葉系以外に、中胚葉系ならびに内胚葉系への分化能を有することを確認した。この結果に関して、筆頭著者として原著論文を投稿し、PLOS ONEに受理された。

また、前年度に引き続き、単離したニッチを用いた網羅的遺伝子発現解析を展開している。問題となっていた細胞溶解液の組成を再検討することで、微量サンプルからのcDNA Libraryの作製に成功した。
