Outline of Annual Research Achievements

本研究は、下垂体幹細胞が外部より侵入している可能性に着目し、遺伝子改変動物を作製・解析する事で下垂体幹細胞が外部より侵入している事を明らかにする事を目的とした。
これまでの我々の解析結果より、下垂体幹・前駆細胞で発現しているPrrx1, Prrx2, Prop1, S100b遺伝子に着目した。それぞれの遺伝子のプロモーター下に、Creリコンビナーゼと改変エストロジェン受容体の融合タンパク質であるCreERT2を結合させ、遺伝子発現状況下でCreERT2を発現する遺伝子改変ラットの作出を試み、Prrx1-CreERT2、S100b-CreERT2ラットを作出した。レポーター遺伝子であるCAG-CAT-Venus配列を遺伝子導入したラットと交配し、Prrx1- or S100b-CreERT2/CAG-CAT-Venusラットを作出した。作出された遺伝子改変ラットでは、CreERT2遺伝子の発現が確認された。しかし、Creに対する特異的な抗体を用いた免疫組織学的解析によるCreERT2の検出を試みたが、陽性シグナルは得られなかった。タモキシフェン投与後にVenusの発現も得られなかった事から、遺伝子改変動物の作出に失敗したと判断した。
同時に、様々な侵入へする事が知られている神経堤細胞に着目し、神経堤細胞と神経堤由来細胞を特異的に標識するP0-Cre/CAG-CAT-EGFPマウスを用いた解析を行なった。その結果、神経堤細胞は下垂体発生初期に下垂体に侵入し、すぐに下垂体幹細胞へと分化転換している事が明らかとなった。また、成体下垂体では全ての下垂体ホルモン産生細胞の一部へと分化している事に加え、一部の神経堤細胞は未分化性を有したまま局在している事を明らかにした。
この事から、胎仔期に外部より侵入した神経堤細胞が下垂体幹細胞としてホルモン産生細胞を供給している事が明らかとなった。
