Outline of Annual Research Achievements

乾燥・凍結に耐性を持つ南極クマムシActuncus antarcticusのゲノム解析を行い、総塩基数28.9 Gbの配列を得た。これにより完成したミトコンドリアの全長配列(14.4 kb)の解析結果を、国際的科学雑誌Mitochondoriaに投稿するべく、現在、論文の準備を進めている。一方、本来の目的であったクマムシの核ゲノム配列については、細菌ゲノム配列の混入のために完成には至らなかった。そこでクマムシの無菌化を行い、餌のクロレラとの二者培養の系を確立した。これによりクマムシの高純度の長鎖ゲノムDNAを調整することが可能になり、10x genomics社のchromium、並びにPacBio社のRSIIによる完全長ゲノムの決定を開始する準備が整った。
さらに、南極線虫Panagrolaimus davidiから得られた乾燥・凍結耐性遺伝子の候補遺伝子LEAを、実験モデル生物Caenorhabditis elegansに遺伝子導入し、本来、乾燥感受性の生物であるC. elegansに乾燥耐性を付与することに成功した。これは耐性遺伝子の実用的な応用に大きなインパクトを持つ結果だと考えている。
また、乾燥耐性との強い関連を持つ高温耐性生物（温泉に生息する）について、2報の論文を発表した。
以上のように、数々の困難を乗り越え、研究のは好ましい方向に向かっていたが、残念ながら研究を続けるポストに就くことができず、科研費申請資格を喪失し、本研究を中止することになった。成果を期待され研究費を交付された研究を全う出来ず、慙愧の念に堪えない。現在、これまでに整備した実験材料や研究成果を活かせるよう、これまでの共同研究者と調整を行っている。
