Outline of Final Research Achievements |
The nuclear pore complex has an important role in various biological natures, such as nuclear form maintenance, the nucleo-cytoplasmic transport and nuclear morphology assembly, but it is suggested that the function of the nuclear pore complex is not necessarily limited to them. It is strongly suggested that Tmem48 null mice phenotypes. Tmem48 is one of nucleoporins, which are the constituent building blocks of the nuclear pore complex. Tmem48 null mice show abnormalities of both the axial skeleton and gametogenesis, although Tmem48 is ubiquitous expression. In this study, it is proposed to elucidate the role of the nucleoporin in vivo by nucleroprin reporter mice, which expressed the fusion gene of Tmem48 and an antibody tag protein or a fluorescence protein by using the genome editing technique, such as CRISPR/Cas9 system.
