Outline of Final Research Achievements

The researcher evaluated MAO-A and MAO-B activity of 48 cathinone derivatives using a commercial kit. Except for 4-methoxy-N,N-dimethylcathinone (4-MeO-DMC), all tested cathinones did not show significant MAO-A inhibition activity. In addition, all tested cathinones (including 4-MeO-DMC) did not show significant MAO-B inhibition activity. IC50 of 4-MeO-DMC for MAO-A inhibition activity was 63.1 uM. This activity was equivalent to 1/13 of 4-methoxyamphetamine, 1/4 of 4-methoxymethamphetamine, and 1.2 times of 4-methoxyethylamphetamine.

Academic Significance and Societal Importance of the Research Achievements

カチノン類の中枢興奮作用は、文献的には、「シナプス前細胞へのモノアミン類の再取り込み阻害」、「シナプス小胞からのモノアミン類の放出促進」、「モノアミンオキシダーゼ(MAO)の阻害」の3種類の機序によると考えられてきた。しかしながら、カチノン類のMAO阻害作用を網羅的に検討した研究は報告されてこなかった。本研究において、4-メトキシ-N,N-ジメチルカチノンを除き、有意なMAO阻害作用を有するカチノン類は認められなかったことから、カチノン類全般の中枢興奮作用は、モノアミン類の再取り込み阻害作用あるいはモノアミン類の放出促進が主たるものであることが明らかとなった。
