Outline of Final Research Achievements |
The short hairpin RNA (shRNA) library is often used to detect promising candidates for specific molecular target therapies in cancer. For our study, we first performed a shRNA library analysis to identify cellular viability-related genes in breast cancer TNBC and non-TNBC cell lines. Then, we performed a transcriptome analysis ton select highly expressed genes in TNBC compared with non-TNBC cell lines. Finally, we used transcriptome analysis data from the public database RefEx to select the specific genes not expressed in normal organs. From these analyses, we identified four genes as candidates for molecular target therapy in TNBC, one is Caspase-14 and the other is Carboxypeptidase A4. We performed several immunohistochemical analysis, using tissue microarray from breast cancer human surgical specimens and migration assay and proliferation assay, using cell lines. We had several significant data.
