Outline of Annual Research Achievements

平成28年度は、昨年度に引き続き、顔面肩甲上腕型筋ジストロフィー患者の遺伝学的解析を行った。浸透率が低い、または症状に差異のあるFSHD1の家系においては、FSHD2の原因として知られるSMCHD1に変異を持ち、FSHD1においても症状を修飾する可能性のある家系が存在することを見いだした。FSHD1家系においても、引き続き新規疾患修飾因子の検索を行う必要があることが分かった。
研究年度全体を通して、FSHD発症に関連する、D4Z4にエピジェネティックな影響を与える因子の探索を行うとともに、FSHD患者300例以上の解析を行った。これまでに知られているSMCHD1に加えて、LRIF1やDNMT3Bなどの新規の遺伝子が、疾患発症に関わっている可能性を見いだした。さらなる新しい因子の発見にむけて、D4Z4結合タンパク質の網羅的解析に取り組んだが、enChiP法による免疫沈降法は、非特異的結合が多く、解析には至らなかった。今後、さらに方法の改良が必要と考えられた。
