Outline of Annual Research Achievements

平成27年度に引き続き、平成28年度においてもコリン性蕁麻疹における汗アレルギーの研究を行った。
①Balb/cマウスのフットパッドに対してアセトン処理、またはテープストリッピングを施行し、角層にダメージを与えた状態で、ピキア酵母で作成した組換えMGL_1304を反復塗布して、凍結切片を作成、蛍光ラベルしたタグに対する抗体でMGL_1304の検出を試みた。アセトン処理したマウスフットパッドにおいて、組換えMGL_1304が真皮に著明に浸透している像が確認できた。しかしながら、MGL_1304が汗腺に特に取りこまれている像は認めなかった。
②MGL_1304以外のマラセチア蛋白(MGL_1781)が、ヒト蛋白(TRX: thioredoxin)と交差すること、またTRXが表皮や汗腺に局在することを明らかにした。コリン性蕁麻疹患者血清は、ウェスタンブロットでTRXと弱い結合は示すものの、明らかなヒスタミン遊離は示さなかった。TRXはマラセチアに過敏性を示すアトピー性皮膚炎の病態に関与している可能性はあるが、Ⅰ型アレルギー反応に関与している可能性は低いと考えた。
③MGL_1304と構造的に相同性のある蛋白をクローニングし、組換え蛋白を作製したところ、アトピー性皮膚炎患者血清IgEと結合することが明らかになった。また、MGL_1304特異的モノクローナル抗体もこの蛋白を認識していた。さらにこの組換え蛋白は弱いヒスタミン遊離活性を有していることも明らかになった。
